You want it to pressurize the intake manifold, because often that is the biggest problem area. And boost will have no problem getting into the intake manifold with the TB closed. If air couldn't get past the TB plate when throttle is closed, then the car wouldn't be able to idle. If you put 30 psi of pressure into your turbo inlet and you don't see any boost on your gauge, then you have a major leak or 20.
